Montego Air Flight 828 took off from Jamaica on the night of April 7, 2013. After three hours and nineteen minutes in the air, an otherwise routine flight punctuated by an encounter with a "sudden weather surge," Flight 828 landed in New York – on the evening of November 4, 2018. The plane had been missing, and all aboard presumed dead, for five years, six months, and twenty-eight days.

Manifest follows the passengers and crew who skipped through time, and their loved ones who have been living without them, as they try to rebuild their lives. As they adjust to their "new normal," the passengers of Flight 828 begin to realize that whatever force it was that propelled them into the future is calling them to some larger purpose.
